چکیده

Field A: Source planting of all transgenic apple plants expressing sense/antisense S6PDH. This planting was already in existence at the start of this project and contains all of the source materials of different transgenic clones planted in a randomized design on their own roots at a 15x15 ft spacing. The plot contains 354 apple trees of which 53 are control trees of the golden delicious apple cultivars, Greensleeves (GS; 26) and Ginger Gold (GG; 27). The other trees are transgenic apple trees of which 106 trees express an anti-S6PDH, 111 trees express a sense-S6PDH and the remainding 74 trees express other constructs not relevant to this present study.

Silencing leaf sorbitol synthesis alters long-distance partitioning and apple fruit quality.

Sorbitol and sucrose are major products of photosynthesis distributed in apple trees (Malus domestica Borkh. cv. "Greensleeves") that affect quality in fruit. Transgenic apple plants were silenced or up-regulated for sorbitol-6-phosphate dehydrogenase by using the CaMV35S promoter to define the role of sorbitol distribution in fruit development. Transgenic plants with suppressed sorbitol-6-phos...

Nucleotide Sequence of a cDNA Encoding NADP-Sorbitol-6-Phosphate Dehydrogenase from Apple.

Sorbitol plays a key role in the translocation of photosynthate in the Rosaceae family, including apple (Malus domestica), pear, and stone fruits (4, 7). NADP-S6PDH2 plays the most important role in biosynthesis of sorbitol in apple because its activity increases before accumulation of sorbitol in seedlings (8), and high activity is maintained in leaves (10). This enzyme reduces G6P to S6P usin...

Ubiquitous distribution and different subcellular localization of sorbitol dehydrogenase in fruit and leaf of apple

NAD(+)-dependent sorbitol dehydrogenase (NAD-SDH, EC 1.1.1.14), a key enzyme in sorbitol metabolism, plays an important role in regulating sink strength and determining the quality of apple fruit. Understanding the tissue and subcellular localization of NAD-SDH is helpful for understanding sorbitol metabolism in the apple. In this study, two NAD-SDH cDNA sequences were isolated from apple fruit...

Purification and characterization of sorbitol-6-phosphate phosphatase from apple leaves

Sorbitol-6-phosphate phosphatase (SorPP; EC 3.1.3.50) catalyzes the final step in sorbitol biosynthesis in sorbitol-synthesizing plant species, but its kinetic and regulatory properties have not been characterized. In this study, the enzyme was purified 1727-fold to apparent homogeneity from apple leaves with a maximal specific activity of 89.8 mmol min 1 mg 1 protein measured at 2 mM sorbitol-...

Down-regulation of sorbitol dehydrogenase and up-regulation of sucrose synthase in shoot tips of the transgenic apple trees with decreased sorbitol synthesis.

Both sorbitol and sucrose are translocated to, and utilized in, sink tissues of apple (Malus domestica). Considering that antisense suppression of aldose 6-phosphate reductase resulted in lower concentrations of sorbitol and higher concentrations of sucrose in source leaves without altering the vegetative growth of apple trees, it was hypothesized that sorbitol metabolism is down-regulated and ...
